By selecting the volume of the injected sample, the volume of the flow channeland the flow rates of streams at the confluence points, dispersion can be adjusted to a medium value. Systems with medium dispersion are suitable for automation of all reagent-based assays.
If sensitivity of the assay should be maximized,
- the flow rate should be reduced (pFI)
- sample volume should be increased so that the dispersion coefficient is adjusted to D 2-4.
- reagents should be added through a confluence points.
- concentration of reagents should be increased.
- the temperature of the system should be elevated.
- a long path flow cell should be used.
